📋 Clinical Overview

Polyethylene Glycol 3350 (PEG 3350) is a high-molecular-weight, non-absorbable, osmotic laxative. In the Indian context, it is a first-line therapy for the treatment of functional constipation and for bowel preparation prior to colonoscopy or colorectal surgery. It works by retaining water in the intestinal lumen via osmotic forces, softening the stool, increasing stool volume, and stimulating peristalsis without causing significant electrolyte imbalance. The 8.5gm sachet is a standard adult dose for constipation management.

💊 Dosage & Administration

Adult: For constipation: 8.5gm (one sachet) dissolved in 150-200 mL of water, once daily. Dose may be adjusted based on response (range: 8.5gm to 34gm per day in divided doses). For bowel preparation: As per specific bowel prep kit instructions (e.g., 2-4 sachets in divided doses with large volumes of clear fluids).

Note: Empty the entire contents of the sachet into a glass. Add 150-200 mL of cool water (or clear liquid like juice without pulp), stir until completely dissolved, and drink immediately. Do not use hot water. It is best taken once daily, at any convenient time. For bowel prep, follow a clear liquid diet and specific timing instructions provided by the physician.

🔬 Mechanism of Action

Polyethylene Glycol 3350 is a high-molecular-weight polymer that acts as an osmotic agent. After oral administration, it dissolves in water and is not absorbed from the gastrointestinal tract. It creates an osmotic gradient that draws and retains water within the colonic lumen. This increase in intraluminal water content hydrates and softens the stool, increases stool volume, and stimulates colonic motility through mechanical distension, leading to defecation.
