📋 Clinical Overview

A fixed-dose combination (FDC) of a cholinergic precursor (Alpha-GPC) and a nootropic racetam (Piracetam) used primarily for cognitive enhancement and management of cognitive deficits. Alpha-GPC increases acetylcholine synthesis, while Piracetam modulates neurotransmission and neuronal membrane fluidity, providing a synergistic effect on cognitive functions like memory, attention, and learning. This combination is widely used in India for age-related cognitive decline and post-stroke recovery.

💊 Dosage & Administration

Adult: One tablet (containing Alpha-GPC 250mg + Piracetam 800mg) twice daily, preferably with meals. May be initiated at once daily for tolerance.

Note: Administer orally with a full glass of water, preferably with food to minimize gastrointestinal side effects. Do not crush or chew unless advised. Maintain adequate hydration.

⚠️ Contraindications

  • Hypersensitivity to Piracetam, Alpha-GPC, or any component
  • Patients with cerebral hemorrhage (acute phase)
  • Severe renal impairment (eGFR <30 mL/min) due to piracetam accumulation
  • Huntington's chorea (piracetam may exacerbate symptoms)

🔬 Mechanism of Action

The combination works synergistically. Alpha-GPC is a highly bioavailable source of choline, crossing the blood-brain barrier to increase the synthesis of the neurotransmitter acetylcholine. Piracetam modulates neuronal and vascular functions, improving cerebral microcirculation, neuronal membrane fluidity, and facilitating interhemispheric communication via the corpus callosum. It may also potentiate cholinergic neurotransmission and protect against hypoxia.

🤰 Special Populations

Pregnancy: Category B (Piracetam). Animal studies show no risk, but human data is insufficient. Alpha-GPC safety is not established. Use only if potential benefit justifies potential risk to the fetus. Not recommended.

Driving: May cause dizziness, drowsiness, or blurred vision, especially at treatment initiation. Patients should not drive or operate machinery until their individual response is known.
