A fixed-dose combination topical medication for psoriasis, combining a vitamin D3 analogue (calcipotriol) with a super-potent topical corticosteroid (clobetasol propionate). This combination provides synergistic anti-psoriatic, anti-inflammatory, and immunosuppressive effects, offering superior efficacy and faster onset of action compared to monotherapy, while potentially reducing the cumulative steroid exposure and associated side effects.
Adult: Apply a thin layer once or twice daily to the affected areas. Total weekly dose should not exceed 100 grams. Treatment should be limited to 4 weeks. After a 4-week course, a treatment-free interval is recommended.
Note: Wash hands before and after application. Apply a thin layer and rub gently until absorbed. Do not occlude the area with bandages or dressings unless directed by a physician. Avoid contact with eyes, mouth, and mucous membranes. Do not use on broken or infected skin. For scalp psoriasis, part hair and apply directly to lesions.
The combination exerts a synergistic effect on psoriasis via complementary pathways. Calcipotriol modulates keratinocyte differentiation and proliferation by binding to the vitamin D receptor (VDR), while clobetasol provides potent anti-inflammatory, immunosuppressive, and antiproliferative effects by activating glucocorticoid receptors (GR).
Pregnancy: Category C (US FDA). Topical corticosteroids, especially potent ones, may pose a risk to the fetus if used in large amounts or for prolonged periods. Use only if potential benefit justifies potential risk to the fetus. Avoid extensive use, occlusion, or high doses.
Driving: No known effects on driving ability.
