📋 Clinical Overview

Adapalene is a third-generation synthetic topical retinoid-like compound, structurally related to retinoic acid. It is a selective modulator of retinoic acid receptors (RAR-β and RAR-γ). It is a mainstay in the topical treatment of mild to moderate acne vulgaris in the Indian population, valued for its efficacy, favorable tolerability profile compared to older retinoids, and photostability.

💊 Dosage & Administration

Adult: Apply a thin film once daily, in the evening, to clean, dry affected areas of the skin.

Note: Wash face with a mild, non-medicated cleanser and pat dry. Wait 20-30 minutes for skin to dry completely. Apply a pea-sized amount for the entire face, avoiding eyes, lips, nostrils, and mucous membranes. Use fingertips to spread gently. Do not apply to unaffected skin. Wash hands after application.

🔬 Mechanism of Action

Adapalene binds to specific nuclear retinoic acid receptors (RAR-β and RAR-γ) within keratinocytes and sebocytes. This binding modulates gene transcription, leading to normalization of follicular keratinization (reducing microcomedone formation), anti-inflammatory effects, and comedolysis.

🤰 Special Populations

Pregnancy: Category C. Animal studies have shown fetal abnormalities (retinoid effects). No adequate and well-controlled studies in pregnant women. Use only if the potential benefit justifies the potential risk to the fetus. Topical application results in minimal systemic absorption, but caution is advised.

Driving: No effect. A topical agent with no CNS activity.
