Price: ₹160 - ₹300 (MRP)
Mfr: Macleods Pharmaceuticals Ltd. | Form: Topical Gel

📋 Clinical Overview

A topical fixed-dose combination gel/cream primarily indicated for the treatment of mild to moderate inflammatory acne vulgaris. It combines the antibacterial action of clindamycin, the anti-inflammatory and sebum-regulating properties of niacinamide, and the keratolytic and wound-healing effects of allantoin. This combination targets multiple pathogenic factors of acne: bacterial proliferation, inflammation, excess sebum, and abnormal follicular keratinization.

💊 Dosage & Administration

Adult: Apply a thin film to the affected area(s) twice daily, morning and evening, after cleansing and drying the skin.

Note: 1. Wash hands and affected area with a gentle cleanser. Pat dry. 2. Apply a pea-sized amount for the entire face (or proportionally for smaller areas). 3. Gently spread as a thin, even film. 4. Avoid eyes, lips, nostrils, mucous membranes, and broken skin. 5. May be used under non-comedogenic moisturizer/sunscreen if needed.

⚠️ Contraindications

  • History of hypersensitivity to clindamycin, lincomycin, niacinamide, allantoin, or any component of the formulation
  • History of regional enteritis, ulcerative colitis, or antibiotic-associated colitis

🔬 Mechanism of Action

The combination exerts a multi-targeted effect on acne pathogenesis. Clindamycin inhibits protein synthesis in Cutibacterium acnes, reducing bacterial colonization and inflammatory mediators. Niacinamide exerts anti-inflammatory effects by inhibiting leukocyte chemotaxis and the release of pro-inflammatory cytokines (IL-1, TNF-α). It also reduces sebum production and improves skin barrier function. Allantoin promotes keratolysis (softening and shedding of the stratum corneum), unclogs pores, and stimulates epithelial cell proliferation and wound healing.

🤕 Side Effects

  • Dryness of skin
  • Mild erythema (redness)
  • Peeling/flaking at application site
  • Mild burning or stinging sensation (usually transient)

🤰 Special Populations

Pregnancy: Category B (US FDA). Clindamycin topical is considered low risk. Niacinamide and allantoin are not expected to pose a risk. However, use only if clearly needed and under medical supervision. Large-scale human studies are lacking.

Driving: No effect on driving ability.

🔄 Drug Interactions

Erythromycin (topical or systemic)Antagonism; both bind to the same ribosomal site, reducing clindamycin's antibacterial efficacy.Major
Neuromuscular blocking agents (e.g., Pancuronium)Clindamycin may enhance neuromuscular blockade.Moderate (relevant only if significant systemic absorption)
Topical Retinoids (Tretinoin, Adapalene)Increased risk of skin irritation, dryness, and peeling. Recommend staggered application (e.g., one in AM, one in PM).Moderate
Topical Benzoyl PeroxidePotential for mutual inactivation if applied simultaneously. Also increases irritation risk. Can be used at different times of day.Moderate
